UV Cabinet Calibration in Pharmaceuticals

Learn how to calibrate the UV Cabinet used to view the spots on Thin Layer Chromatographic (TLC) plate.

For the lamp having wavelength (Emission) at about 254 nm

1. Select a pre-coated thin layer chromatographic plate coated with silica gel G as coating substance.
2. Prepare a solution of Sodium salicylate in methanol having a concentration of 0.04%w/v.
3. The weight of Sodium Salicylate 40.0 mg in methanol 100 ml.
4. Apply 5mL of this solution on the plate.
5. Dry in the air and place it in the UV chamber.
6. Select the lamp having a wavelength (emission) of about 254 nm.
7. Check for the compliance.
8. The lamp complies if a clear spot of about 5 mm diameter is revealed.

For the lamp having wavelength (Emission) at about 365 nm

1. Select a pre-coated thin layer chromatographic plate coated with silica gel G as coating substance.
2. Prepare a solution of Sodium salicylate in methanol having a concentration of 0.2%w/v.
3. The weight of Sodium Salicylate 200.0 mg in methanol 100 ml.
4. Apply 5mL of this solution on the plate.
5. Dry in the air and place it in the UV chamber.
6. Select the lamp having a wavelength (emission) of about 365 nm.
7. Check for the compliance.
8. The lamp complies if a clear spot of about 5 mm diameter is revealed.
